Epistasis plays an important role in the genetic architecture of common human diseases and can be viewed from two perspectives, biological and statistical, each derived from and leading to different assumptions and research strategies. Biological epistasis is the result of physical interactions among biomolecules within gene regulatory networks and biochemical pathways in an individual such that the effect of a gene on a phenotype is dependent on one or more other genes. In contrast, statistical epistasis is defined as deviation from (...) additivity in a mathematical model summarizing the relationship between multilocus genotypes and phenotypic variation in a population. The goal of this essay is to review definitions and examples of biological and statistical epistasis and to explore the relationship between the two. Specifically, we present and discuss the following two questions in the context of human health and disease. First, when does statistical evidence of epistasis in human populations imply underlying biomolecular interactions in the etiology of disease? Second, when do biomolecular interactions produce patterns of statistical epistasis in human populations?Answers to these two reciprocal questions will provide an important framework for using genetic information to improve our ability to diagnose, prevent and treat common human diseases. We propose that systems biology will provide the necessary information for addressing these questions and that model systems such as bacteria, yeast and digital organisms will be a useful place to start. We are told that we live in a scientific world. All about us are the fruits of scientific research, and the products of scientific industry. But, in spite of this transformation of our material surroundings, scientific thought, or the scientific method as such, has had no effect upon the everyday thought and behaviour of our people. To be sure, along with the scientific gadgets a few scientific truths have been disseminated. They know the earth is round and moves about the (...) sun; they could know that bacteria cause disease and that they are themselves descended from less articulate forms of animal life. Even when such conclusions are accepted, it is on the basis of authority with no understanding as to how they arose and on what basis they stand. For the large majority, science has merely acquired a cloak of respectability; it is a stronger magic than that of our forebears, and our modern alchemy is known as a powerful though mysterious source of pecuniary profit. Religion is intrinsically social, and hence irretrievably organizational, although organization is often seen as the darker side of the religious experience--power, routinization, and bureaucracy. Religion and secular organizations have long received separate scholarly scrutiny, but until now their confluence has been little considered. This interdisciplinary collection of mostly unpublished papers is the first volume to remedy the deficit. The project grew out of a three-year inquiry into religious institutions undertaken by Yale University's Program on Non-Profit Organizations and sponsored by the (...) Lilly Endowment. The scholars who took part in this effort weree challenged to apply new perspectives to the study of religious organizations, especially that strand of contemporary secular organizational theory known as "New Institutionalism." The result was this groundbreaking volume, which includes papers on various aspects of such topics as the historical sources and patterns of U.S. religious organizations, contemporary patterns of denominational authority, the congregation as an organization, and the interface between religious and secular institutions and movements. With what right and with what meaning does Heidegger use the term 'truth' to characterize Dasein's disclosedness? This is the question at the focal point of Ernst Tugendhat's long-standing critique of Heidegger's understanding of truth, one to which he finds no answer in Heidegger's treatment of truth in §44 of Being and Time or his later work. To put the question differently: insofar as unconcealment or disclosedness is normally understood as the condition for the possibility of propositional truth rather than (...) truth itself, what does it mean to say - as Heidegger does - that disclosedness is the "primordial phenomenon of truth" and what justifies that claim? The central aim of this paper is to show that Tugendhat's critique remains unanswered. Recent Heidegger scholarship, though it confronts Tugendhat, has not produced a viable answer to his criticism, in part because it overlooks his basic question and therefore misconstrues the thrust of his objections. Ultimately, the paper suggests that what is needed is a re-evaluation of Heidegger's analysis of truth in light of a more accurate understanding of Tugendhat's critique. The paper concludes by sketching the profile of a more satisfactory reply to Tugendhat's critical question, advocating a return to Heidegger's 'existential' analyses in Being and Time in order to locate the normative resources Tugendhat finds lacking in Heidegger's concept of truth. In the past two decades, reinforcement learning has become a popular framework for understanding brain function. A key component of RL models, prediction error, has been associated with neural signals throughout the brain, including subcortical nuclei, primary sensory cortices, and prefrontal cortex. Depending on the location in which activity is observed, the functional interpretation of prediction error may change: Prediction errors may reflect a discrepancy in the anticipated and actual value of reward, a signal indicating the salience or novelty of (...) a stimulus, and many other interpretations. Anterior cingulate cortex has long been recognized as a region involved in processing behavioral error, and recent computational models of the region have expanded this interpretation to include a more general role for the region in predicting likely events, broadly construed, and signaling deviations between expected and observed events. Ongoing modeling work investigating the interaction between ACC and additional regions involved in cognitive control suggests an even broader role for cingulate in computing a hierarchically structured surprise signal critical for learning models of the environment. The result is a predictive coding model of the frontal lobes, suggesting that predictive coding may be a unifying computational principle across the neocortex. (shrink)

The pedagogical technique of the playful Plato, especially his ability to create living discourses that directly address the student, is the subject of Plato the Teacher. “The crisis of the Republic” refers to the decisive moment in his central dialogue when philosopher-readers realize that Plato’s is challenging them to choose justice by going back down into the dangerous Cave of political life for the sake of the greater Good, as both Socrates and Cicero did.

As scientific and engineering efforts become increasingly global in nature, the need to understand differences in perceptions of research ethics issues across countries and cultures is imperative. However, investigations into the connection between nationality and ethical decision-making in the sciences have largely generated mixed results. In Study 1 of this paper, a measure of biases and compensatory strategies that could influence ethical decisions was administered. Results from this study indicated that graduate students from the United States and international graduate students (...) studying in the US are prone to different biases. Based on these findings, recommendations are made for developing ethics education interventions to target these decision-making biases. In Study 2, we employed an ethics training intervention based on ethical sensemaking and used a well-established measure of ethical decision-making that more fully captures the content of ethical judgment. Similar to Study 1, the results obtained in this study suggest differences do exist between graduate students from the US and international graduate students in ethical decision-making prior to taking the research ethics training. However, similar effects were observed for both groups following the completion of the ethics training intervention. (shrink)

In this book, William H. F. Altman argues that it is not order of composition but reading order that makes Euthyphro, Apology of Socrates, Crito, and Phaedo “late dialogues,” and shows why Plato’s decision to interpolate the notoriously “late” Sophist and Statesman between Euthyphro and Apology deserves more respect from interpreters.

In the first section, I consider what several logicians say informally about the notion of logical consequence. There is significant variation among these accounts, they are sometimes poorly explained, and some of them are clearly at odds with the usual technical definition. In the second section, I first argue that a certain kind of informal account—one that includes elements of necessity, generality, and apriority—is approximately correct. Next I refine this account and consider several important questions about it, including the appropriate (...) characterization of necessity, the criterion for selecting logical constants, and the exact role of apriority. I argue, among other things, that there is no need to recognize a special logical sense of necessity and that the selection of terms to serve as logical constants is ultimately a pragmatic matter. In the third section, I consider whether the informal account I have presented and defended is adequately represented by the usual technical definition. I show that it is, and provably so, for certain limited ways of selecting logical constants. In the general case, however, there seems to be no way to be sure that the technical and informal accounts coincide. (shrink)
